Argireline
Cosmetic and Skincare
ABOUT
Argireline is a topical cosmetic hexapeptide marketed as a needle-free alternative to botulinum toxin. Its skin penetration is the weak point: over 99% of an applied dose does not reach the dermis.
EVIDENCE OVERVIEW
Too little published work to judge either way
WHAT THIS MEANS
Insufficient evidence. Review the research and limitations before relying on this record.
IDENTITY
HALF-LIFE
HOW IT IS THOUGHT TO WORK
A synthetic acetylated hexapeptide patterned on the N-terminal end of SNAP-25. It competes with SNAP-25 for binding to the vesicle-associated membrane protein, destabilising ternary SNARE complex formation and inhibiting neuronal exocytosis. That is a genuinely different mechanism from botulinum toxin, which is injected and enzymatically cleaves SNAP-25 to produce irreversible blockade for three to four months - argireline competes reversibly and only where it arrives.
PHARMACOKINETICS
This is the central problem. At roughly 889 daltons and highly charged and hydrophilic, it does not readily cross the lipophilic stratum corneum by passive diffusion. A 2015 in-vitro penetration study found over 99% of applied argireline failed to reach the dermis, with only 0.01% detected in the viable epidermis. Whether meaningful concentrations reach neuromuscular junctions is unresolved.

HUMAN EVIDENCE
Small and inconsistent. The available base comprises a 60-participant randomised trial, a 19-participant randomised trial, and two lower-certainty studies. The strongest positive result (Wang 2013) was manufacturer-funded; an independent 2023 evaluation was null. One 92-participant study tested it in combination with other actives, so benefit cannot be attributed to the peptide. Older claims of up to 48% efficacy after four weeks of twice-daily use are not supported by independent summaries, which put realistic dynamic-line reductions around 10-30% in small, often industry-sponsored samples. A plausible alternative explanation for observed benefit is hydration and film-forming effects of the vehicle.
REPORTED EFFECTS
Acute toxicity is negligible - on the order of 2000 mg/kg, against 20 ng/kg for botulinum toxin. A 2020 uncontrolled study of a 10% gelcream in 26 patients reported improvements in hydration, elasticity and sebum with no allergic reactions; it contributes to the safety rather than the efficacy literature. It is regulated by FDA as a cosmetic ingredient, not a drug, with no IND or NDA filed and no approved therapeutic indication. Marketing it as "topical Botox" overstates what the penetration data supports.
RESEARCH LIMITATIONS
This is a source-backed summary, not a recommendation. Read the mechanism, pharmacokinetic, human-evidence and reported-effect fields separately: a mechanism is not an outcome. Availability, legal status and product quality differ by jurisdiction; consult a qualified clinician.
