Kisspeptin-10
Hormonal and Signaling
ABOUT
Kisspeptin-10 is the decapeptide fragment of kisspeptin that activates KISS1R to trigger GnRH release. It has been studied in humans as a reproductive-axis probe and potential IVF trigger, but is not approved anywhere.

IDENTITY
HALF-LIFE
HOW IT IS THOUGHT TO WORK
Kisspeptin-10 is the C-terminal decapeptide of the KISS1 gene product and the endogenous agonist for KISS1R (GPR54). It acts upstream of GnRH: kisspeptin stimulates GnRH secretion from hypothalamic GnRH neurons expressing GPR54, which in turn drives pituitary LH and FSH release. Because it works through the physiological GnRH pulse generator rather than bypassing it, the LH response depends on an intact hypothalamic-pituitary axis.
PHARMACOKINETICS
Kisspeptin-10 is cleared within minutes, giving a sharp, short LH response to bolus dosing. Kisspeptin-54 has a longer duration of action, which is why trials needing sustained stimulation have generally used the 54-residue form. Precise human half-life values are not curated here.
REVIEW
PUBLISHED PAIR RECORDS
with Gonadorelin
Sequential steps in the same reproductive axisKisspeptin-10 and gonadorelin act at different points on one pathway rather than on two. Kisspeptin acts upstream, on KISS1R-expressing GnRH neurons, to drive GnRH release; gonadorelin is GnRH itself, acting directly on pituitary gonadotrophs. Giving both does not recruit a second mechanism - it stimulates the same axis at two consecutive points, and the downstream step is the shared bottleneck. Dose pattern matters more than dose for both: pulsatile GnRH stimulates gonadotropin release while continuous or supraphysiological exposure downregulates the receptor and suppresses output, and gonadotropin responses to kisspeptin have been shown to desensitise under sustained exposure in animal work. A schedule that produces continuous rather than pulsatile stimulation can therefore invert the intended effect. Peppy reports curated pharmacology, not a safety verdict. A prescriber or pharmacist should review any intended combination.
Reviewed 21 Aug 2026HUMAN EVIDENCE
Intravenous bolus kisspeptin-10 produces a rapid, dose-dependent rise in serum LH, with maximal stimulation at 1 microgram per kilogram in healthy men. In men with congenital hypogonadotropic hypogonadism who had undergone reversal, exogenous kisspeptin elicited a GnRH-induced LH pulse, demonstrating the axis was intact. These are small mechanistic studies - typically four to six subjects - conducted in research settings, not efficacy trials. Kisspeptin-54 has been studied as an IVF trigger. No kisspeptin product is approved in any jurisdiction.
REPORTED EFFECTS
The short-duration studies reported no significant safety concerns, but sample sizes were very small and exposure brief. Continuous exposure to kisspeptin causes receptor desensitisation, the same phenomenon that turns continuous GnRH agonism into gonadal suppression - so a sustained-dosing schedule cannot be assumed to produce a sustained effect. Kisspeptin-10 was added to FDA's Category 2 interim compounding list in September 2023 and was reviewed at the October 2024 Pharmacy Compounding Advisory Committee meeting, where FDA recommended against inclusion in the 503A Bulks Regulation.
RESEARCH LIMITATIONS
This is a source-backed summary, not a recommendation. Read the mechanism, pharmacokinetic, human-evidence and reported-effect fields separately: a mechanism is not an outcome. Availability, legal status and product quality differ by jurisdiction; consult a qualified clinician.
